As part of this work, Biomarker Ltd. developed a culture medium for the in vitro viability assay of primary human tumor cells called OmniSanguine.
It is difficult to study primary human tumor cells in vitro. Although there are a large number of in vitro adapted tumor cell lines, most tumor cells, when removed from the human body, do not survive. There are various defined cell culture media that are optimized for isolation of different types of primary cells. These media contain extra additives and growth factors that are specifically selected to meet the needs of one given cell type at a time and they are not interchangable.
OmniSanguine is the first broad spectrum tissue culture medium that supports the in vitro survival of a large variety of primary human (tumor) cells.
OmniSanguine overcomes the limitations of the conventional defined media by closely mimicking the growth supporting conditions of the human blood. OmniSanguine was developed at the Karolinska Institutet, Stockholm, Sweden. OmniSanguine was specifically optimized to support the survival of a wide array of human tumor cells in vitro in order to allow short term viability assays using a large panel of anti-cancer agents. OmniSanguine was succesfully used to assess the effect of different licensed and experimental drugs on freshly isolated tumor cells from the blood or bone marrow of patients suffering from:
Acute lymphoid leukemia
Chronic lymphoid leukemia
Various types of acute myeloid leukemia
Mantle cell lymphoma
HHV8 carrying body cavity lymphomas
Immunosuppression associated immunoblastic lymphomas
In addition OmniSanguine was shown to support the assessment of drug sensitivity of tumor cells isolated from body cavity effusions (ascites, pleural fluid) of patients suffering from late stage malignant conditions such as:
carcinomas of the stomach, colon, rectum, ovarium, endometrium, cervix, pancreas, breast, lung as well as malignant mesotheliomas.
