OmniSanguine is a dark particle free fluid with very high protein content that is particularly suited for fluorescence based high throughput viability and high content cell physiological assays using automated cellular imaging. OmniSanguine is particularly suitable for the three days in vitro viability assay of tumor cells (iVV assay) in combination with the fluorescent live/dead discrimantion staining using VitalDye. The dark non-transparent background serves as a perfect optical quenching agent thus allowing the study of the cell physiological effect of anti-cancer drugs with high autofluorescence such as anthracyclines.
OmniSanguine is optimally buffered with carefully controlled physiological ion and redox values. It also satisfies the need of tumor cells for extra glucose and glutamate.
OmniSanguine is prepared with a patented manufacturing procedure.
VitalDye is a fluorescent dye mixture which differentially stains living and dead human or animal cells. Living cells are labelled with a strong green fluorescence whereas the nuclei of dead cells show strong red fluorescence. VitalDye is optimized for standard FITC, Rhodamine filter cubes available on most fluorescent microscopes (Excitation at 488 nm, Emission at 525 nm and excitation at 545 nm, emission at 620nm, respectively). The dye is provided as 100x diluted stock solution that is stable at 4°C for several weeks.
OmniSanguine is delivered in frozen vials on dry ice. It should be preserved at -80°C untill use. Upon thawing, the vials sould be used immediately. Due to the very high protein content, insoluble precipitates may fall out after a few days of culturing at 37 °C. These precipitates have high specific density, have no effect on cell viability and do not adhere to or ingested by any of the cell types tested. The delicate primary cells can be easily recovered from cultures with protein precipitates using conventional ficoll density centrifugation on a Lymphoprep cushion.
